Abstract

Time resolved acquisition of fluorescence provides the advantages of reduced background reflection and reduced analyte autofluorescencee, considerably improving the signal to background ratio. Europium chelate with its relatively longer fluorescence emission decay period can be captured by simple and inexpensive detectors. In this work we have developed a TRF reader using a monochrome camera and its associated exposure triggering circuitry to capture time resolved fluorescence from Europium Chelate striped on nitrocellulose (NC) membrane. The reader is imaging based and does not require complex mechanical movement of the test strip under investigation. Ten different concentrations of Europium between 100 ng/ml to 1000 ng/ml were imaged, and an improvement in the signal to background ratio of up to 70% was observed. The output of the developed TRF reader was found to be linear (R2=0.9873) over the range of concentrations.
