We report a point-of-care platform for rapid detection of Neisseria gonorrhoeae (NG) with integrated nucleic acid purification, amplification, and detection on a disposable thermoplastic cartridge in under 15 minutes. Sample preparation is automated by permanent magnet actuation of functionalized magnetic particles through preloaded reagents within the assay cartridge. On-cartridge PCR thermocycling and fluorescence measurements allows duplexed detection of two genetic NG targets for pathogen identification and assessment of susceptibility to antibiotics (ciprofloxacin). The assay was evaluated using 44 clinical isolates and swabs with >97% sensitivity and 100% specificity.
